CCR5基因多态性的研究进展

　　2．3  CCR5m303  CCR5m303是指在CCR5读码框内303位碱基由A→T的点突变。突变使读码框内形成一个终止子，导致CCR5的翻译提前完成而产生无功能的CCR5穿膜蛋白，其作用与CCR5纯合突变一样也有抵抗HIV1感染的作用。如果同一个体内2条染色体同时发生CCR5△32和CCR5m303突变，将导致CCR5功能的全部丧失，这些个体对CCR5依赖病毒有着极强的抵抗力［14］。CCR5m303在人群中的突变率较低，Quillent等人报道意大利人为0．2％［14］，而我国尚未见个例报道［15］。 ——内容来自www.17lw.com,一起论文网

　　2．4  CCR5894C  CCR5894C是指CCR5基因编码区内894位C碱基的缺失而导致后面相邻的299308位氨基酸发生改变，使随后的44个氨基酸由于翻译终止而缺失，最后造成CCR5受体蛋白的C末端的胞内部分发生改变，对CCR5的磷酸化及信号的传导功能产生一定的影响［16,17］。Ansari等报道CCR5894C为中国及日本人所特有［16］。刘明旭等通过对汉族和部分少数民族的调查，仅发现汉族(2．15％)及藏族(0．53％)存在CCR5894C，且高危人群的HIV阴、阳个体之间的突变频率没有明显的相关性，因此，他们认为CCR5894C在中国人群中对HIV1的易感性没有太大的影响［18］。3  小结 ——防采集设置www.17lw.com,一起论文网

　　随着我国对AIDS预防力度的加大，经性途径传播方式感染HIV的人数所占的比重逐渐增加。因此，CCR5基因多态性的研究为AIDS的预防及治疗提供了新的手段和方法。CCR5调控区、非编码区为多态性区域，而编码区的多态性频率较低，但编码区基因多态性研究仍取得了重大进展并不断有新的突变被发现。目前公认的能对HIV感染和AIDS进程有影响的只有CCR5△32和CCR5m303，而CCR5894C的作用仍不十分明确，这些多态性存在着明显的地域和人种间的差异［10］。我国是多民族国家，因此我国少数民族CCR5基因多态性的普查，尤其是一些国人特有的基因多态性的分析，将对我国AIDS的预防和治疗有一定的指导意义。 ——论文大聚合www.17lw.com,一起论文网
